褪黑激素(Melatonin,MT)又称为N-乙酰基-5-甲氧基色胺,在松果体中产生,是调节昼夜节律的主要成分。褪黑激素是由色氨酸通过一系列羟基化和甲基化步骤产生的。褪黑素在肝脏中代谢成6-羟基硫酸褪黑素形式后随尿液排出体外。褪黑素合成后,储存在松果体内,交感神经兴奋支配松果体细胞释放褪黑素。褪黑素的分泌具有明显的昼夜节律,白天分泌受抑制,晚上分泌活跃。褪黑素可抑制下丘脑-垂体-性腺轴,使促性腺激素释放激素、促性腺激素、黄体生成素以及卵泡雌激素的含量均减低,并可直接作用于性腺,降低雄激素、雌激素及孕激素的含量。褪黑激素及其代谢产物具有抗氧化和清除自由基的潜力。还有证据表明,它们在活性氧蛋白途径的调节中发挥作用。研究表明,褪黑激素水平与某些癌症的发病率可能存在相关性。唾液可以用来检测褪黑激素水平。与血清或血浆相比,唾液相对容易获得,并且不含有血清和血浆中存在的诸多干扰物质。唾液中的褪黑激素代表了全身循环的水平,尽管确切的比例尚未完全量化,公布的数值范围为血清值的20-50%。褪黑素在癌症、睡眠周期、衰老和其他领域的作用也越来越引起人们的关注,尤其关注褪黑素受体和循环褪黑素的生物利用度,以及褪黑素作为膳食补充剂的使用。
