最佳缓冲区W？（10毫米TrIS HCl（pH为8.5°C，25°C）；10 mMgCl 2；100 mM NaCl；1毫米DTT）。

最佳温度37°C

储存条件10毫米TrIS HCl（pH 7.5）；250 mM NaCl；0.1毫米EDTA；7毫米2-巯基乙醇；50%甘油；储存在-20°C。

5倍的酶消化后，结扎约20%的DN**段可以结扎。在10%的PEG结扎的情况下是更好的。

在37℃下，用1μgλDNA和10μA酶孵育16小时后，检测到非特异性Hyrasists无非特异性的活性。

重叠坝甲基化阻断的甲基化效应（GMATC）：TCGCGATC。

在80℃下孵育20分钟，使失活酶失活。
