对λDNA（HydiIII消化）的测定

最佳缓冲液Y（33毫米醋酸三乙酯（pH为7.9，在25°C）；10毫米醋酸镁；66毫米醋酸钾；1毫米DTT）。

最佳温度37°C

储存条件10毫米KH2PO4（pH 7.4）；50毫米氯化钾；0.1毫米乙二胺四乙酸；7毫米2-巯基乙醇；200μg/ml牛血清白蛋白；50%甘油。存储在-20°C。

用高浓度的T4DNA连接酶在10%倍PEG存在下，用DNA约50%的DN**段进行2倍过量消化后结扎。其中90%以上可以再剪除。

在37℃下，用1μgλDNA和30μA酶孵育16小时后，检测到非特异性Hyrasists无非特异性的活性。

甲基化敏感性未经测试的甲基化效应

在80℃下孵育20分钟，使失活酶失活。
