Immunogen: Phosphotyramine-KLH Reactivity: All species expected Product Description: Anti-phosphotyrosine antibody (clone 3B6) was developed by using phosphotyramine-KLH conjugates as immunogen. This antibody can be used in western blotting to examine the modification of tyrosine phosphorylation in immunoprecipitated proteins or the overall profile of tyrosine phosphorylation in cells and tissues. Source/Purification: The mouse monoclonal antibody is produced by immunizing animals with synthetic phosp-Tyr-containing peptides and immunoaffinity purified by protein G. Specificity: Anti-phosphotyrosine mouse mAb (clone 3B6) is a high affinity antibody selectively against phosphorylated tyrosine. ELISA and dot blot assay using a wide variety of phospho-peptides indicate that this antibody only binds to tyrosine-phosphorylated peptides independent of the surrounding amino acid sequence but not to threonine, serine or unmodified peptides. Western blotting analysis on crude proteins extracted from HeLa cells shows that this antibody interacts with a broad range of tyrosine-phosphorylated proteins induced by sodium vanadate treatment. Recommended Applications: ELISA, WB, IP *WB=Western blotting; IP=Immunopreciptation. Recommended dilution: WB:1:1000, IP: 1:200. IMPORTANT: For Western blotting, incubate membrane with diluted antibody in 5% BSA, 1 x TBS, 0.1% Tween-20 at 4℃ with gentle shaking, overnight. Use at an assay dependent concentration.Optimal dilutions/concentrations should be determined by the end user and maybe assay dependent. Not yet tested in other applications. | ||
Scientific Description: Tyrosine phosphorylation is one of the major means of cell signaling transduction in multiple pathophysiological conditions regulated by tyrosine kinase activities. In cancer, unregulated tyrosine kinase activity can drive tumor malignancy and formation by generating inappropriate proliferation and survival signals. Antibodies that specifically recognize phosphorylated tyrosine residues have proved to be invaluable to these studies.
