分子量:72 kDa

纯度:>92% as determined by SDS-PAGE.

内毒素:Less than 1.0 EU per μg of the Human PCSK9 (D374Y), His Tag by the LAL method.

Buffer:PBS, pH7.4

生物活性:Measured by its binding ability in a functional ELISA. Immobilized Human LDL R, His Tag (Cat# LDR-H5224) at 10 μg/mL (100 μl/well) can bind Human PCSK9 (D374Y), His Tag (Cat# PCY-H5225 ) with a linear range of 5-50 ng/mL.

产品特性:Human PCSK9 (D374Y) Protein is fused with a polyhistidine tag at the C-terminus, and has a calculated MW of 72 kDa. The predicted N-terminus is Gln 31. The reducing (R) protein migrates as 18 kDa and 65 kDa in SDS-PAGE due to glycosylation and proteolytic digestion. The D374Y mutation results in higher affinity of PCSK9 for LDLR.

产品背景:Proprotein convertase subtilisin/kexin type 9 (PCSK9), is an enzyme which in humans is encoded by the PCSK9 gene. This gene encodes a proprotein convertase belonging to the proteinase K subfamily of the secretory subtilase family. This protein plays a major regulatory role in cholesterol homeostasis. PCSK9 binds to the epidermal growth factor-like repeat A (EGF-A) domain of the low-density lipoprotein receptor (LDLR), inducing LDLR degradation. PCSK9 may also have a role in the differentiation of cortical neurons. Mutations in this gene have been associated with a rare form of autosomal dominant familial hypercholesterolemia (HCHOLA3).Especially,the D374Y mutation results in higher affinity of PCSK9 for LDLR.
